Chronic hyperglycaemia associated with diabetes mellitus (DM) can caused reproductive problems in men, suggested due to hormonal changes, neuropathy, and oxidative stress pathway. Vitamin D, such as cholecalciferol, is one of antioxidant agent that known can improve hyperglycaemia in DM. However, the effect of vitamin D on infertility is still requires further investigation. The aim of this study is to elucidate the effect of cholecalciferol on epithelium thickness of seminiferous tubules in diabetic male rats. Nineteen adult male rats (Rattus norvegicus) of Wistar strain were induced became DM with combination high fat diet for 3 weeks and a single intra peritoneal injection of low dose streptozotocin (STZ) (35 mg/kg BW) at the end of second week. Animals then were divided into control group (K), and 3 treatment groups (X1, X2, and X3), that were administered cholecalciferol per oral with doses 6.25 μg/kg BW, 12.5 μg/kg BW, 25 μg/kg BW, respectively, for 14 days. Testes were removed and processed into histological preparation with Haematoxylin Eosin staining. Epithelium thickness of seminiferous tubules was measured with 10x of objective lens magnification, using Image Raster 3 software. Data were analyzed with one way ANOVA test (α= 0.05, significancy p<0.05). Epithelium thickness of seminiferous tubules were 72.12±6.31 μm in group K, 78.34±6.42 μm in group X1, 104.31±9.89 μm in group X2, and 117.52±9.23 μm in group X3. One way ANOVA test showed significant difference in epithelium thickness of seminiferous tubule among groups (p=0.000). Administration of cholecalciferol with different doses can improve epithelium thickness of seminiferous tubules in diabetic male rats.
